The cofactor (NADPH) was essential for the enzyme activity, and the oxidase activity was activated by Cu2+ or by addition of cytochrome c. The oxygen evolving activity of glucose dehydrogenase was inhibited by cyanide. The molar ratio of reduced nicotinamide cofactors to the oxidase-active protein was close to 1.0 in the soluble fraction of the cells. Under the optimum conditions, the enzyme catalyzed the conversion of 2.5-8.0 mM CO into 25-100% CO2. The presence of nitrite, N2O, cyanide and other inhibitors had a very little effect on the rate of CO conversion. The carbon monoxide dehydrogenase was present in the soluble and membrane fractions of the cells.
